Acetaminophen, a commonly used analgesic (pain reliever) and antipyretic (fever reducer) was analyzed. It is a challenging compound to run by mass spectrometry as a result of its low molecular weight (151) and fragility. It was run in the positive ion mode by electrospray using MRM mode monitoring of 152/110. The HPLC separation was accomplished at the flow rate of 200ul/min by employing 25/37.5/37.5 water/acetonitrile/methanol with 0.1% formic acid using a Beta Basic C18 2.1 X 50 mm 3micron column injecting 5ul. The signal to noise, based on 3 sigma, was 36/1 on the 50 fg/ul injection. The concentration range run shows excellent linearity. The transition 609 to195 was monitored using MRM. The signal to noise is better then 90:1. Here a 5 microliter injection volume is used. The chromatographic conditions were 3/7 water/Acetonitrile 2mM ammonium acetate with 0.1% formic acid at a flow of 0.2 milliliter per minute. MS Method Developer The MolAna MS method developer arranges each of your MS methods into three basic sections: method, experiment, and mass tables. Since the advanced design and electronics on the 3Q allows for virtually every MS parameter to be changed within a single run, method section essentially sets the overall time and quadrupole calibrations settings, unlike previous generation triple quads in which the polarity or source type had to be fixed for the entire run. MolAna allows for multiple overlapping experiments to be performed on any given sample enabling the researcher to get the maximum amount of information from the smallest amount of sample. Both the mass table section and the experiment section are completely interfaced with MolAna’s Data Processing system so that you can easily build acquisition methods from the interpretation of previously analyzed samples.
